Domestic serum typically has shorter lead times than South American serum, with many products available from stock or shippable within 3-7 days, but you need to confirm whether it is in stock or scheduled for production. For large-volume purchases, it is advisable to ask the supplier to lock the lot number or provide retention samples to avoid batch changes mid-order that could affect experiments. In addition, serum requires cold-chain handling; a short lead time does not guarantee safety. Upon arrival, check temperature records and packaging integrity, and perform sterility and mycoplasma spot testing. A price of 10,000 yuan/L is considered mid-to-high range for domestic serum, so it is worth requesting more complete quality documentation from the supplier, not just a COA.

On the COA, endotoxin ≤10 EU/mL and hemoglobin ≤20 mg/dL are considered moderate levels for routine cell culture, not particularly stringent. Mesenchymal stem cells are sensitive to serum lot-to-lot variation, and poor late-stage cell status is not necessarily directly caused by these two parameters alone; other unlisted items such as hormones, growth factors, and exosomes must also be considered. It is recommended to perform parallel passaging comparisons between this lot and the previous lot, focusing on doubling time, attachment rate, and phenotypic markers. If the parallel results are poor, then investigate items beyond the COA, such as biochemical values and oxidative stress indicators. These two parameters alone, being within range, cannot fully explain the cell status.

Domestic fetal bovine serum at around 10,000 RMB/L can indeed be found from several brands whose COAs show endotoxin and hemoglobin levels close to the imported lot you have on hand, but a COA is the supplier's self-testing, so you can't rely on the paperwork alone. I suggest first requesting COAs from 3–5 consecutive lots from the domestic manufacturer to check lot-to-lot variation in endotoxin and hemoglobin; then apply for a 50 mL trial sample and run a side-by-side culture of the same MSC batch against your current South American serum for at least 3 passages, recording morphology, viability, and trilineage differentiation potential. Once validation is OK, then discuss bulk purchasing, and have the supplier specify acceptance criteria and return conditions in the contract.
