用量2L，如果买10L大包装液体，开封后保存是个问题。液体PBS开封后容易长菌，不建议整桶反复抽取；可先分装成500 mL或1 L无菌瓶，但普通养殖场分装很难保证无菌。干粉包装更耐放，但要防潮，开封后扎紧密封。从成本看10L液体单价可能更低，但要确认自己能否在一个月内用完；否则粉剂小包装更稳。

渗透压和离子浓度方面，稀释用PBS应接近等渗，一般在280-310 mOsm/kg；磷酸盐浓度常用0.01 M左右，氯化钠约0.85%。过高或过低都可能刺激鸡群鼻黏膜或改变病毒存活环境。买粉剂后最好用纯化水按说明配，定容准确，别随意估计水量。如果有条件，配好后用渗透压仪测一下，至少每批抽测。

细胞培养级PBS能否直接给鸡用，要看成分表。多数细胞培养级PBS无钙镁、无菌、内毒素低，理论上可以作为一般稀释用，但如果含有酚红、EDTA或抗凝剂等额外成分，就不建议直接用于疫苗稀释。养殖场建议优先用疫苗厂家推荐的专用稀释液；如果必须替代，找无酚红、无EDTA的PBS，并先做小群试验观察应激和接种反应。

pH偏碱确实可能影响疫苗中活病毒的稳定性。禽用活疫苗稀释后应尽快使用，稀释液pH一般建议在7.0-7.4，偏碱尤其超过7.8时可能加快病毒失活。配完PBS后要用校准过的pH计测，偏碱可用少量盐酸或换用酸性配方微调，不要凭试纸估算。若效价持续不稳，先排除稀释液pH和现配现用时间。
