Clumping is indeed not just a culture medium issue; rotation speed, inoculation density, and flask type have a major impact. For a 125 ml shake flask, I typically use 25-30 ml working volume at 110-130 rpm, and the orbital diameter must be adjusted accordingly. Inoculation density should be 0.3-0.5×10^6/ml; too low or too high a density can both lead to clumping. During passaging, do not pipette too vigorously; use a gentle pipette to mix as much as possible to reduce DNA release. If clumping starts from the third passage, also check whether the incubator CO2 is stable and whether the flask cap is too tight. You can first fix the process and repeat one batch with the old medium to confirm it is not operational drift, then switch to the new medium.

When selecting an alternative, it is recommended that candidate manufacturers provide publicly available application data for Expi293 or HEK293 suspension culture, CoA, and animal-origin-free statements. Prioritize options with matching feed and transfection workflows, as this will save considerable effort for transient transfection later. Before ordering, confirm the shelf life and inventory of small packaging such as 500 ml to avoid overstocking. For validation, three groups can be set up: old medium control, direct replacement with new medium, and gradient adaptation to new medium. In addition to viability, also record cell diameter, aggregation rate, and protein titer. Also test a batch under transfection conditions, because changes in medium may affect PEI/liposome transfection efficiency. Keep a backup of the old medium; do not replace everything all at once.

Clumping often appears in the later stage of the adaptation period. Previously, with imported media, it only became obvious by the third passage, possibly related to cell metabolites, calcium and magnesium concentrations, or the feeding strategy. When switching to domestic media, gradient adaptation is recommended: new:old = 25:75 for one passage, 50:50 for one passage, and 75:25 for another passage, observing viability, doubling time, and clumping rate for 48 h at each passage. Do not look only at viability; counting the proportion of aggregates with >4 cells under the microscope is more meaningful. If the new medium has higher calcium and magnesium, clumping may be more likely; you can ask the manufacturer for the formulation range or whether it is suitable for 293F. During evaluation, also assess target protein expression at the same time; do not pursue only good cell growth.

If using Expi293F, it is recommended to directly approach domestic brands with extensive experience in HEK293 suspension platforms, such as OPM, Bioengine, GenScript, and Yuanpei, and first request 50–100 mL or 125 mL shake flask trial samples. Clarify whether they offer serum-free, animal origin-free versions adapted for Expi293/HEK293, whether they contain anti-shear components such as PF-68, and whether the matching feed is in stock. For small-scale use at 2–3 × 125 mL per week, it is advisable to first perform a parallel recovery: preserve the culture in the old medium, subculture twice at 1:1 in the new medium, and monitor viability and doubling time. In terms of lead time, domestic products generally arrive within 1–2 weeks, but availability of small-pack sizes must be confirmed to avoid receiving near-expiry samples.
