Another easily overlooked aspect is Raw264.7 passage number, serum starvation, and lysis buffer. If the Raw264.7 passage number is too high or the culture is too dense, the cells will become desensitized. I keep the cells within P8, plate them at 60-70% density, and then starve them with 0.5% FBS for 2-4 hours, which gives lower background than without starvation. Add sufficient phosphatase inhibitors to the lysis buffer, preferably using a formulation containing NaF and Na3VO4, and do not over-wash after membrane transfer. Before IL-6 stimulation, the cells can be washed twice with plain medium. When endotoxin is suspected, add polymyxin B in parallel or reconstitute with low-endotoxin water to rule out LPS interference.

I suggest switching your detection readout from Western blot to a more sensitive flow-based phosphorylation assay or cell lysate ELISA for initial screening, especially when p-STAT3 is weak, as Western blot has a narrow dynamic window. If you insist on Western blot, choose CST 9145 as the primary antibody, a rabbit monoclonal antibody, incubate overnight at 4°C, keep the secondary antibody dilution not too high, and use gradient exposure during development. Regarding brands, ask suppliers for small samples for comparison: use the same plate of cells, the same concentration gradient, and run old batch, new batch, and different brands together; this is more reliable than asking the community about a single subjective experience. Imported brands are stable but expensive and have long lead times; domestic brands are suitable for high-frequency use, provided you stock up on multiple units of the same lot number at once.

I mainly use domestic products now, focusing on ED50 and batch reports. Before purchasing, ask the manufacturer to provide the CoA for that lot and check the specific activity (ED50) and purity, not just the milligram quantity. After arrival, run a p-STAT3 dose curve using your Raw264.7 cells, keep the sample preparation and exposure conditions fixed, and use an old lot for bridging; if the new lot only shows bands at 50 ng/ml, you can directly request a return or exchange or use it for non-critical experiments. Regarding lead time, many domestic products are in stock and can arrive in 3-5 days; imported agents often take 2-4 weeks. In terms of price, domestic 50 μg is roughly several hundred to over a thousand, while imported products are more expensive, and you need to inquire for specific pricing.

Don't just focus on the brand first—rule out endotoxin and handling issues first. Raw264.7 is extremely sensitive to LPS. If IL-6 has high endotoxin levels, it can cause background phosphorylation or alter the inflammatory state, which may actually narrow the gradient window. I suggest you ask the supplier for the endotoxin data for that lot, and prioritize lots below 0.1 EU/μg, carrier-free or low-endotoxin. After reconstitution, aliquot into low-protein-binding tubes to avoid repeated freeze-thaw cycles; at the same time, test 0.1, 1, 10, 50, and 100 ng/ml at two time points, 15 and 30 minutes—don't just focus on 15 minutes. I've used imported PeproTech/R&D, which have relatively stable lot-to-lot activity; domestic Nearshore and Sino Biological also have mouse IL-6, but it's best to run your own parallel titration on a lot before using it.
