The price gap between domestic and imported products mainly lies in endotoxin quality control, activity calibration, and batch-to-batch consistency. For signaling pathway studies, purity is not critical, but endotoxin cannot be overlooked: at 20 ng/mL stimulation, elevated endotoxin levels can interfere with results through pathways such as TLR4/NF-κB. It is recommended that domestic products also be required to have endotoxin levels stated on the COA.

Whether activity declines over long-term storage depends largely on preservation and shipping. Recombinant TNF-α is not recommended to undergo repeated freeze-thaw cycles. After dissolving the lyophilized powder, it is best to aliquot according to the amount needed for a single experiment, supplement with 0.1% BSA as a carrier, and store at -80°C. For liquid formulations, reduce the number of times the vial is opened and try to use it within a short period after opening. During shipping, confirm that the cold chain records are complete and avoid leaving it at room temperature in summer. For domestic products, if the manufacturing date is recent, shipping is stable, and the buffer design is reasonable, long-term activity loss is not necessarily worse than that of imported products; the same batch of cells can be used periodically to perform a quality control stimulation point.

Buffer formulation is also critical; for cell experiments, avoid formulations containing detergents or high salt concentrations that are not clearly disclosed. Before ordering, ask clearly about the buffer composition, whether it is sterile-filtered, and whether it contains carrier proteins. If a domestic quote is 800 but the COA is incomplete, it is advisable to first purchase the smallest package, self-test endotoxin with an LAL kit, and compare dose curves with an imported lot on the same plate. If endotoxin and activity validation pass, using the domestic product in signaling pathway experiments is entirely acceptable; later, based on usage requirements, ask the supplier to fix the lot number or reserve samples in advance.

For batch-to-batch stability, it is recommended to establish your own simplified verification rather than relying solely on the manufacturer's COA. After receipt, use the same cell passage for concentration-gradient stimulation, for example 0.2, 1, 5, 20, and 50 ng/mL, with fixed stimulation time and downstream readouts (such as phosphorylated protein or target gene), and fit the EC50. Use the first batch as the baseline, and compare subsequent batches on the same plate upon arrival. If the EC50 drifts by more than 2-3 fold, attention should be raised. This is more relevant to your own experimental system than simply trusting the ED50, and it also allows you to quickly determine whether the problem is protein batch variation or a change in cell state.
