There are actually quite a few domestic manufacturers making 10x-compatible droplet chips, in Suzhou, Hangzhou, and Shenzhen, with lead times typically 5-10 working days, shorter than your three weeks. But don't just take the salesperson's word for compatibility. Be sure to ask clearly whether the chip inlet/outlet dimensions and channel depth match 10x standard consumables. It's best to run a no-load droplet test with the same batch of reagents and check droplet diameter and uniformity. There are options under 150 yuan, but I suggest buying 5-10 pieces as a small sample first, not 20 pieces right away. Regarding batch-to-batch consistency, ask them to provide dimensional inspection data for that batch, then test the clogging rate yourself using fluorescent microspheres or cell suspension, with particular attention to whether the inlet filtration structure is adequate.

The probability of chip clogging is strongly related to the chip inlet filtration design and sample pretreatment, and it is not necessarily all due to domestic products. In my experience with domestic chips, if the inlet has a filter structure of >=40 um, clogging is somewhat better, but the filter itself also increases back pressure. I suggest that after receiving the goods, you first do three things: inspect the channel under a microscope for debris or bonding adhesive overflow; run cell-free buffer for 10 min to see whether the pressure is stable; then run once more with labeled microspheres or nuclei resuspended in PBS, and record the clogging rate. It is best to keep 1-2 pieces per lot as retention samples, so that if a certain lot is found to be no good, you can ask the manufacturer for a replacement. At below 150 per piece, lot-to-lot fluctuation will still exist, and you cannot use the psychological expectation of imported stability.

10x-compatible chips generally only provide the droplet generation structure, while the oil phase and gel beads still need to be matched. If the channel depth differs too much between chips from different manufacturers, using the original 10x oil phase may cause droplet size deviation. It is recommended to first ask the manufacturer whether they have a recommended or validated oil phase formulation, or directly ask whether they have run tests with the original 10x reagents. Some domestic solutions use their own oil phase or lysis buffer, so do not mix and match and then try to troubleshoot the cause. In addition, during operation, pay attention to the compatibility between the oil phase and the chip material. PDMS chips will swell, and if it is a plastic chip, ask about oil resistance. Before purchasing, ask the manufacturer to provide a simple protocol, including flow rate, loading volume, and recommended reagents, which can save a lot of troubleshooting time.
