The background height cannot be solely determined by the brand, as the packaging source, clone number, and verification application are crucial. Aladdin small size packaging is cheaper, but batch repeatability may not be as stable as original large packaging. It is recommended to inquire before purchasing whether the same host/clone number is available, whether there are ICC/IF data or references, and preferably obtain dilution recommendations and staining charts for the batch. After arrival, take the validated primary antibody from our laboratory for parallel staining: same batch of cell slides, same fixed permeability conditions, compare signal-to-noise ratio. When the background is high, optimization can be based on the blocking solution (5% BSA or 10% serum), secondary antibody dilution, washing solution Triton concentration, and washing times. There is no need to directly determine that the antibody is not effective. The delivery time is usually 3-7 days, but there are significant fluctuations in inventory and logistics. Please ask the sales team to check the spot goods and confirm the cold chain in advance.

From a procurement risk perspective, cold chain stability in second-tier cities and the storage condition of aliquoted antibodies deserve more attention than price. When placing an order, note: "Requires cold chain, include temperature recording card, avoid room-temperature transfer." Upon receipt, check whether sufficient ice packs are used inside the packaging and whether the antibody tubes have condensation but no large ice crystals. If 100 μL aliquots undergo repeated freeze-thaw cycles, the titer drops significantly. It is recommended to aliquot into 5-10 μL per tube upon arrival and store at -20/-80, avoiding repeated use of the entire tube. This specification is acceptable for preliminary experiments or a small number of samples. If subsequent critical data are needed, it is recommended to use validated imported antibodies or purchase a larger size from the same lot. High background may not be solely an antibody issue; cell fixation, excessive permeabilization, and nonspecific secondary antibody binding should all be investigated. Therefore, run system controls before drawing conclusions.

Aladdin sub-packaged a few non-key indicators that I have used, feeling that the specificity varies from target to target, and it cannot be generally said that the background is high. When the expression of the target protein is not high, it is easier to cover the weak signal in the background. Therefore, it is recommended to do an antibody dilution gradient (such as 1:50, 1: 100, 1: 200) first, and run a control with no one antibody and only two antibodies to confirm whether the background is from first antibody or second antibody/blockade. 100 μL diluted with 50-80 μL of working fluid per tablet after 1: 100, theoretically more than ten to twenty tablets can be dyed, but the operation loss and climbing area will reduce the actual number of tablets. The cold chain is transported to second-tier cities, and Aladdin generally uses ice bags and foam boxes, but in the summer, note the temperature record and the number of ice bags, and check whether the inner packaging is close to room temperature after arrival. Not recommended for critical experiments if ice packs are fully developed upon receipt.
