When selecting a third-party provider, beyond qualifications, you should also check whether they have experience serving clients with overseas submissions, and whether their reports can be provided in both Chinese and English, with support for method principles and raw data. Domestic pharmacopeial methods are generally acceptable for NMPA, but during FDA on-site inspections, they often look at data integrity, method validation, and the COA chain, rather than merely recognizing the standard name. It is recommended that the third party specify the limit of detection and reference standards in the report, and if necessary, add comparative data using the 9CFR method. If you only test for BVDV, BPV, and Reo-3, it may be sufficient domestically, but for overseas INDs, it is usually recommended to add parvovirus, especially when you have subsequent filtration steps, as you need to demonstrate the raw material viral load. Retention samples and subcontracting should also be clearly stated in the contract in advance.

Domestic pharmacopoeia methods are usually accepted during NMPA on-site inspections, but the FDA will pay more attention to whether the method source, validation data, and limit of detection can cover your risk assessment. It is recommended not to focus only on the phrase "according to the pharmacopoeia"; require the third party to include the test items, method numbers, cell lines, blind passage numbers, positive controls, and limit of detection in the report. If only the Chinese Pharmacopoeia is followed, overseas IND submissions may require supplementary method comparison against 9 CFR 113.53 or ICH Q5A, or at least a method equivalence statement. My approach is to choose the same institution for incoming inspection to issue reports according to both standards at the same time. The cost will increase, but it can reduce a lot of explanation during the submission stage.

Testing only for BVDV, BPV, and Reo-3 is insufficient for overseas INDs. Especially when FBS is used for cell banks and cell culture, audits often follow up on parvovirus (PPV), PI3, BRSV, and adenovirus. Parvovirus particles are small and heat-resistant, and once they enter downstream purification validation, the situation becomes complicated. Therefore, I recommend including BVDV, BPV, Reo-3, PI3, BRSV, adenovirus, and parvovirus in incoming material testing. If the budget is limited, at minimum do not omit BVDV and parvovirus; other items can be supplemented based on supplier COA risk. Domestic pharmacopeial methods are acceptable, but you must judge the test coverage yourself and cannot simply meet the minimum list.

It depends on the filing region. The FDA gives more recognition to USDA- or EMA-recognized laboratories, while domestic reports often require supplementary documentation.

It is best to find an institution with CNAS and OECD GLP accreditation, and to inquire with the foreign drug regulatory authority in advance.
