For standards, it is recommended not to cut corners. It is best to use the standards provided with the kit or from the same lot, and dilute them with your PBS buffer. Do not directly substitute with water or other buffers. It is recommended to perform spike recovery: add a known concentration of biotin standard to blank PBS and measure it on the same plate as the samples; if the recovery deviates significantly, it indicates matrix interference or that the standard curve is not applicable. Additionally, set up a background control with unlabeled antibody plus PBS to confirm that the antibody itself does not produce false positives. For the limit of detection, do not rely solely on the promotional page. Ask the sales representative for the COA for that lot or the LoD/LoQ in the instructions, as different lots may vary.

Before purchasing, inform technical support of your sample type, buffer, antibody concentration, and whether you are measuring free biotin, and ask clearly whether the kit measures total biotin or can directly measure free biotin, and whether acid hydrolysis or protease digestion is required. For lead time and pricing, it is advisable to request quotes from at least two authorized distributors and ask for the electronic instructions and COA to confirm storage conditions and shelf life; regular imported in-stock items may arrive in a few days, while less common batches may take several weeks, subject to written confirmation from the sales representative. You can also ask the manufacturer whether a small-size trial pack is available, and run a preliminary experiment with your actual samples first to avoid buying the wrong full kit.

First, confirm the kit principle. If it is a HABA/avidin colorimetric method, it is usually not sensitive enough for measuring free biotin, and antibodies in PBS may interfere with the readout; it is recommended to prioritize a fluorescence method based on streptavidin competition or a kit compatible with low concentrations of free biotin. More critically, sample pretreatment: your 200 μL, 2 mg/mL antibody is not recommended for direct loading. First pass it through a Zeba desalting column or 10 kDa ultrafiltration centrifugation to separate unbound free biotin from the labeled antibody, then measure the filtrate/flow-through. Without a separation step, the measured result is difficult to interpret as the proportion of free biotin.
