When choosing ready-to-use internal reference antibodies, I prioritize dilution ratio and shelf life rather than ranking solely by sales volume. The more reliably reputed brands are Proteintech, CST, and Abclonal, but most CST products are not ready-to-use and require self-dilution and aliquoting, with high storage requirements. Ready-to-use products from Proteintech and Abclonal generally contain preservatives and are fine for one year at 4°C; after opening, avoid contamination and repeated warming. During procurement, ask the sales representative to provide the recommended dilution ratio and shelf-life statement for that lot, and preferably request a trial size or small specification. For HeLa/293T, a total protein amount of 20 ug is on the high side; first reduce the loading amount and perform gradient dilution. Laboratories running twice a week should fix the antibody lot number, diluent, and exposure parameters to reduce lot-to-lot variability.

For HeLa and 293T, loading 20 µg for internal control, band smearing is not necessarily due to the antibody itself. First, it is recommended to reduce the loading amount to 10-15 µg, and check whether the transfer buffer is fresh, whether blocking is at least 1 hour, and whether the primary antibody incubation time is too long. For ready-to-use, I have used Proteintech's beta-actin for the past six months, diluted 1:5000, stored at 4°C with a shelf life of about one year, and the bands are relatively solid. If you want to switch directly, first apply for a trial sample, and run the same batch of samples on one membrane side by side with the old antibody for comparison. Abclonal also has small packaging, and the dilution ratio can reach 1:10000, suitable for a throughput of twice a week. After confirming that the new antibody has no nonspecific bands, then purchase in bulk.

Over the past six months, I have mainly compared the ready-to-use loading controls from Proteintech, Abclonal, and Beyotime. Proteintech's β-actin is used at 1:5000-1:10000, and its background and tailing are somewhat better than the aliquoted products I used before; Abclonal's GAPDH bands are relatively clean, and trial applications for small-pack sizes are easy to request; Beyotime is price-friendly, but the dilution ratio is slightly lower, and the shelf life requires attention, so keep it at 4°C and avoid repeated taking. For HeLa and 293T, using 20 ug total protein for loading controls is too high and prone to overexposure and tailing; reducing to 10-15 ug or further increasing the primary antibody dilution ratio will be more stable. It is recommended to apply for trial samples from both companies, run the same batch of samples side by side 3 times, and choose the one with better band morphology and lot-to-lot stability, rather than judging only by a single result.
