When I previously switched to domestic reagents, I placed more emphasis on validation with real serum samples rather than parameters in brochures. Low-concentration IL-6 samples are prone to matrix interference. It is recommended to take at least 5 serum samples expected to have low values and test them undiluted and at 2-fold and 4-fold dilutions to see whether the dilution linearity is parallel; if the undiluted measured value is below the lower limit of quantification, be cautious. For intra-assay CV, you can do it yourself: select two concentrations, high and low, on the same plate, run 15-20 replicate wells, and calculate the CV. Generally, mature large domestic manufacturers can achieve within 10%, but do not rely only on promotional claims. Another practical point: before purchasing, ask the manufacturer whether they can provide comparison data with imported kits or reference ranges. If not, keep 3-5 samples yourself and test them simultaneously on imported and domestic kits, then perform Bland-Altman analysis or regression to assess bias, focusing on whether the low-value bias is acceptable.

The gap between domestic and imported products is not as large as it was a few years ago, but IL-6 itself is often at low levels in healthy mouse serum, so don't select based only on the nominal sensitivity. It is advisable to first ask the manufacturer to provide a COA and the standard curve for the corresponding lot, focusing on precision near the lower limit of quantification, not just the linear range. For more than 60 samples, if the budget is limited, you can look for a major domestic manufacturer that supports small-size trial use, and first run a pilot test with 1-2 plates: use 3 low, medium, and high serum samples and the manufacturer's diluent to perform parallel replicate wells, and check the intra-assay CV, spike recovery, and serum matrix dilution linearity. After confirmation, try to complete the entire batch of samples with the same lot of kits to avoid cross-lot variation. A single box within 800 is achievable, but don't choose a small brand without a lot release report just to save a few dozen yuan.

When selecting domestic alternatives, first clarify supplier qualifications and batch management—this is more meaningful than simply looking at the linear range. Ask the supplier several key points: whether the antigen and antibody are produced in-house, whether the coating antibody and detection antibody are fixed from the same batch, whether functional batch testing is performed on finished products before release, and whether inter-batch data from at least 2–3 batches can be provided. Lead time and cold chain also cannot be overlooked; ELISA kits are temperature-sensitive, and unstable cold chain logistics from some smaller brands can affect color development. With a budget under 800, it is advisable to request trial samples from 2–3 suppliers simultaneously, use the same batch of serum on the same plate or on the same day for side-by-side comparison, and examine the standard curve OD dynamic range, blank background, and low-value discrimination. Finally, for more than 60 samples, it is best to purchase enough of the same lot in one go and keep an extra half-plate margin for repeat testing.
