For macrophage inflammation models, endotoxin is more critical than brand. TNF-α itself activates macrophages, and if endotoxin levels are high, it is difficult to distinguish whether the effect is due to TNF-α activity or LPS background. It is recommended to first ask domestic suppliers to provide the endotoxin test report for that batch, preferably below 0.1 EU/μg; if it only states below 1 EU/μg, it is usually acceptable for cell experiments, but an LPS control should be included. The advantage of imported low-endotoxin products lies in consistency, but lead times are long. You can first buy one vial from a domestic supplier for trial and use the same batch to run a complete dose curve.

The batch-to-batch stability of domestic recombinant proteins cannot be generalized; it depends on whether the supplier has a quality control system. Prioritize those with ISO or GMP backgrounds that can provide a COA for every batch; if a vendor dares to claim low endotoxin based on just a single electrophoresis gel image, be cautious. My approach is to buy small-pack sizes and test two batches: one for functional experiments and one to freeze and store as an internal reference standard. Domestic proteins with stable quality can usually meet the requirements for routine cell experiments, but before critical functional experiments, it is advisable to perform another endotoxin and ED50 validation.

If you only use 2–3 vials per month and your budget is not particularly tight, it is advisable to keep one lot each of imported and domestic products. Imported products arrive slowly, but you can stock one vial first as a positive reference; domestic products arrive quickly, so use them for routine preliminary experiments and condition optimization. This way, not all experiments have to wait for the imported product. When ordering, pay attention to the reconstitution instructions: for TNF-α, try to reconstitute according to the instructions and aliquot in small volumes to avoid repeated freeze-thaw cycles; if the activity of the domestic product declines after reconstitution, the advantage in lead time is meaningless.

Activity data cannot be taken at face value just because they claim activity. Imported products generally provide ED50 or EC50 data, such as L929 cytotoxicity or NF-κB reporter gene data; if domestic products do not provide complete data, you can request the batch COA from the sales representative, especially SDS-PAGE, SEC-HPLC, and activity assay results. Once obtained, run a dose curve yourself using your existing cells, with a low dose range of 0.1–100 ng/mL, and observe inflammatory markers. If there is significant batch-to-batch variation, try to use the same vial for the same series of experiments to avoid switching products midway.

For lead time and budget, choosing domestic products is acceptable, but acceptance testing must be made a fixed procedure. Upon arrival, first check the appearance, COA, and lot number, then reconstitute according to the instructions and test endotoxin and functional indicators. For endotoxin, you can use limulus amebocyte lysate or use cells with polymyxin B for rapid judgment; for example, if IL-6 or TNF expression decreases after adding polymyxin B, it indicates an endotoxin background. If a domestic lot passes, then increase procurement; if the data from two consecutive lots are unstable, switch suppliers. Using imported products for key paper data and domestic products for experimental optimization is a common compromise.
