For intracellular IL-17A staining after Th17 differentiation, it is recommended to perform surface staining for CD4 before fixation and permeabilization, and then stain for IL-17A after fixation and permeabilization. Many protocols are designed this way to avoid the impact of fixation on the CD4 epitope and to clearly gate CD4+ T cells. Surface CD4 should first be stained with a fluorescent label before fixation, washed, and then fixed and permeabilized using Foxp3/Transcription Factor Buffer or BD Cytofix/Cytoperm, with the IL-17A antibody stained in Perm/Wash buffer. This allows the existing surface CD4 antibody to continue being used without the need to purchase an additional intracellular CD4 antibody.

If the budget is tight, for IL-17A you can consider cost-effective domestic brands such as Dakewe, 4A Biotech, MultiSciences, Elabscience, etc., but be sure to ask clearly whether they have been validated for mouse intracellular IL-17A staining, and it is best to have reference images of PMA/ionomycin-stimulated splenocytes. Classic clone numbers such as eBio17B7 and TC11-18H10 have more data available among imported brands, and domestic brands may have the same or alternative clones. Before purchasing, ask the distributor to provide the clone number, specifications, and buffer requirements, and do not look only at price. For imported in-stock conventional clones, it is generally 2-4 weeks, and small pack sizes may take longer.

Whether a surface CD4 antibody can be directly used for intracellular staining after fixation and permeabilization depends primarily on whether the clone has been validated for intracellular staining. Fixation and permeabilization can alter the conformation of the CD4 molecule; if the epitope recognized by the surface antibody is cross-linked or masked, staining intensity may decrease significantly, and background may also increase due to nonspecific adsorption caused by the fixative. It is recommended not to use it directly in a formal experiment. First perform a parallel pilot experiment: divide the same mouse splenocytes into two groups, one for surface staining and one for staining after fixation and permeabilization, and compare the CD4-positive rate and MFI. If there is a marked decrease or high background, this indicates that the clone is not suitable for intracellular staining.

The cost of intracellular flow cytometry antibodies is not just the unit price of the antibody; it also includes fixation/permeabilization buffer, Perm/Wash buffer, and controls. Among imported brands, Invitrogen/eBioscience's Th17 intracellular staining system is relatively mature, and BioLegend and BD also have corresponding products. You can first ask domestic distributors to quote for BD, BioLegend, and Invitrogen, specifying the clone number, size (25 μg or 100 μg), lead time, and whether buffer is included. If it is urgent, look for domestic stock and request a COA. Some distributors can apply for trial samples or small sizes; doing a pilot experiment before bulk purchasing can reduce risk.

Elevated background after fixation and permeabilization is very common and is not necessarily due to incorrect antibody use. It is recommended to add CD16/32 blocking before intracellular staining; many protocols retain the blocking agent in Perm/Wash buffer, so do not omit it. Washing should be sufficient, and fixation time should follow the instructions, as excessive fixation increases autofluorescence and nonspecific binding. Gating must use FMO or isotype controls; especially when the IL-17A-positive proportion is low, FMO is more reliable than isotype controls. If the background remains high, reduce the antibody concentration, increase the number of washes, or add 1% rat serum to the staining buffer.
