Don't just compare the price per 96T; calculate the cost per sample: with the same 96-well format, if you need to run a standard curve + blank + replicates, the actual number of sample wells will differ, resulting in different per-sample costs. Domestic in-stock products are indeed fast, but it is advisable to first buy 1 box and compare it with a small imported sample using the same samples, especially since lavage fluid sample values may be low. In terms of lead time, 2-3 weeks for imports is not unreasonable, provided the distributor has clear customs clearance and cold chain records; for domestic products claimed to be in stock, ask clearly whether it is a full box or a broken box, and whether the shelf life is fresh. BALF can be tested with a regular standard diluent, but it is best to add a set of BALF matrix spikes to determine recovery.

For measuring TNF-α in rat BALF, buffer issues deserve more attention than lead time. The ionic strength and protein content of BALF differ from those of serum, and some ELISA kits will yield falsely high or low results when samples are measured directly without dilution. It is recommended that after receiving the kit, test two dilution levels using pooled BALF and select the dilution factor that falls within the middle range of the standard curve; if sample volume is sufficient, preparing a separate BALF matrix standard is also an option, but kits generally do not include a dedicated BALF buffer. When purchasing, ask technical support whether the kit supports matrices such as "serum/plasma/culture supernatant/lavage fluid"—do not rely solely on a distributor's verbal assurance that it "can measure" the sample.

We frequently measure rat TNF-α in batches for efficacy studies. Regarding lead time, ordering imports through a distributor normally takes 2-3 weeks; if the distributor has bonded warehouse stock, it may take 3-5 days. Domestic products may be in stock, but that does not mean they are suitable; you should first check the COA and intra-assay CV. It is recommended that you ask the distributor to reserve enough of the same lot number, at least the same lot number for more than 200 samples, and additionally request one extra box for backup or repeat testing. When storing BALF samples, centrifuge to remove cells, aliquot, and store at -80°C to avoid repeated freeze-thaw cycles; it is best to run separate standard curves for bronchoalveolar lavage fluid and serum, or at least separate QC, because matrix differences can affect background wells.

The unit price of rat TNF-α 96T can differ by one to two times between domestic and imported products. For imported kits, if there is domestic stock, it usually ships within one week; sourcing from overseas typically takes 2-4 weeks, which is normal. For more than 200 samples, it is recommended to calculate the plate layout first: standard curve + blank + replicates. One 96T plate can usually test 30-40 samples, so you may need 4-6 kits. Try to lock in the same lot number. Bronchoalveolar lavage fluid (BALF) has not been validated for all kits, so be sure to ask the distributor for BALF matrix validation or at least run a pilot test. BALF is generally centrifuged and the supernatant is tested directly; it may need to be diluted 1:2 or 1:5 with the kit's standard diluent, depending on the TNF-α level.

For bulk procurement, it is advisable to have two or three suppliers simultaneously provide: unit price for 96T, delivery time, batch-specific COA, and trial sample policy. With over 200 samples, if there is not enough time to wait for imported kits, you can first use domestic trial samples for preliminary testing; if the spike recovery of domestic and imported kits on mixed samples is comparable, domestic kits can be used while retaining the same lot number. A dedicated buffer for BALF is not mandatory, but you need to perform your own matrix blank and spike validation. In addition, clarify whether the 96T format is a pre-coated plate or requires self-coating; domestic products are cheaper sometimes because costs are saved on consumables and standard preparation, and for long-term large-volume use, reproducibility must also be considered.
