If a suitable kit cannot be obtained in a short time, an internal control can be added to the existing magnetic bead method. Before extraction, add a small amount of internal control RNA/DNA to the lysis buffer, and perform dual-channel qPCR to correct for extraction efficiency and amplification inhibition, so that even if recovery fluctuates, the quantitative results are more comparable. The internal control method is not costly, but it requires redesigning primers and probes and verifying that they do not interfere with the target virus. When purchasing, priority can also be given to kits whose instructions reserve wells for internal controls or explicitly support adding internal controls, making subsequent quantitative analysis easier.

In supplier procurement, we focus on lead time, batch-to-batch variation, and trial samples. Domestic magnetic bead-based nucleic acid extraction kits from Tianlong, Sansure, BioGerm, DaAn, etc. are usually in stock with 3-7 day lead times, while imported MagMAX may take 2-4 weeks. Don't just look at the absolute Ct value; instead, check the CV of extraction from the same low-copy sample across different batches. Batches with large batch-to-batch variation are not suitable for routine monitoring no matter how cheap they are. If the cost is within 15 yuan per test, you can negotiate bulk packaging or research payment terms. Be sure to ask whether proteinase K and magnetic beads are included to avoid secondary procurement. It is best to have the manufacturer send trial samples first and perform small-scale validation using your own sample system.

Our lab has compared the MagMAX magnetic bead method, the QIAamp column method, and domestic Tianlong/Sansure magnetic bead methods on serum with low viral load. In our experience with magnetic bead methods, batches with a large lysis volume, an elution volume of 50-60 μL, and pre-loaded deep-well plates give more consistent Ct values for low-copy quality controls; if procedures are not standardized, deviations are easily amplified. I suggest you take a known low-copy quality control panel or quantified pseudovirus and run 6-8 replicates per vendor to assess intra-assay Ct differences and inter-assay variability. Ask sales to provide recovery data at low viral load, not just total nucleic acid yield.

Fluctuations in recovery rates for low-concentration samples are often not due to the kit itself, but to operational differences. The number of freeze-thaw cycles for serum, whether pipetting and mixing are performed after sample addition, insufficient lysis time, elution buffer not pre-warmed, and inadequate magnetic separation time can all have an impact. After we fixed a single operator, the Ct drift of the low-copy quality control was much smaller. Add one known low-copy quality control to each extraction batch; if the Ct deviates by more than 1 cycle, re-extract the entire batch. Pre-warm the elution buffer to 50-60°C, and after adding it, let it stand at room temperature for 2 minutes before magnetic separation, which can improve recovery.

When selecting, focus on whether the lysis buffer completely lyses enveloped viruses, the magnetic bead particle size and surface modification, and the elution volume. Start with 200 μL of serum/plasma. If the elution volume is too large, low-copy templates will be diluted, and Ct values tend to be higher. With a budget of 15 yuan per test, domestic magnetic bead-based 96-test prefilled kits are generally sufficient. Prioritize models that can extract both RNA and DNA, because some viruses are DNA viruses. Also check whether they are compatible with your laboratory's magnetic rack or automated extraction instrument. Deep-well plates and magnetic rod sleeves are best prefilled to reduce cross-contamination.
