Inter-batch variability cannot be judged by brand alone; it must be managed by lot number. In our group, after each new lot arrives, we first test attachment rate, doubling time, and continuous passaging. For primary macrophages, we also measure endotoxin and hemoglobin. Tumor cells are generally not very sensitive to inter-batch variability, but primary cells may fluctuate significantly. Price differences are sometimes reflected in endotoxin, hemoglobin, and screening costs. It is recommended to ask the vendor to provide cell culture validation data or a small sample for that lot, and to consider switching only after confirming that batch-specific retention samples can be kept and the source is traceable.

In terms of lead time, domestic serum generally has shorter in-stock and logistics cycles than imported serum, but smaller brands may also suddenly go out of stock. It is recommended to choose brands with their own production bases or stable overseas blood collection facilities, sign quarterly stocking agreements with regional distributors, and have the distributors reserve 2-3 bottles as safety stock. Imported Australian-origin serum is more affected by quarantine and logistics, so lead time fluctuations tend to be greater; most domestic South American-origin serum can arrive within 1-2 weeks, but popular catalog numbers are occasionally tight as well. Placing orders one month in advance and requesting batch number locking will make things much more stable.

It is recommended not to switch the entire batch at once. Start by obtaining small samples of the same lot from 1–2 domestic brands sourced from South America, and run parallel comparisons with the current Gibco Australian lot in use. Tumor cells are usually more tolerant, whereas for primary macrophages, focus on adherence, morphology, proliferation curves, and polarization-related markers. Recently, the price gap between imported Australian-origin and domestic South America-origin products is often more than twofold, with domestic South America-origin 500 mL generally falling between 1,000 and 2,000 yuan. Be sure to confirm whether the supplier can provide a complete CoA, blood collection source, viral screening, and endotoxin data.
