The volume in microliters to add per million cells cannot be copied as a fixed value; concentrations vary between batches. After purchase, perform a titration first: for 1×10^6 PBMC, add 0.25, 0.5, 1, 2, and 4 μL respectively. After staining, examine the staining index and the background of the negative population, and select the concentration that gives a high staining index without increasing background. Generally, commercial antibodies are effective at 0.25-1 μg per 1×10^6 cells, but some low-concentration aliquots may require 3-5 μL. If budget control is required, the midpoint of the recommended amount can be used first, and 5-10 samples should be run in a preliminary test before batch processing.

Compensation must be performed with single-stained positive control tubes. It is recommended to use compensation beads or the same cells stained with a single color, with each antibody in a separate tube. Do not rely solely on software-based automatic compensation. For a budget of 30 tubes at no more than 80 yuan per tube, consider BioLegend small-pack sizes, domestic aliquots from Dakewe, Sizhengbo, etc., or BD small specifications, but confirm that the clone and fluorescence are in stock. Domestic distributors typically have a lead time of 3-7 days. Specify the clone number, fluorescence, and specification in advance, and request the same lot. Avoid mixing different lots, as compensation will drift.

Panel suggestion: CD3-FITC, CD8-PerCP-Cy5.5, CTLA-4-PE, LAG-3-APC; most four-color machines can run this, and compensation is manageable. CTLA-4 is weakly expressed, so pairing it with the bright fluorophore PE is appropriate; LAG-3 is sensitive with APC. For clones, I have used CD3 SK7, CD8 RPA-T8, CTLA-4 BNI3, LAG-3 11C3C65 or 3DS223H, with no obvious clone conflicts. CTLA-4 surface levels on resting PBMCs are extremely low, so fixation and permeabilization are required to stain total protein; otherwise, false negatives may occur.

In the workflow, Fc blocking must be performed first, using Human TruStain FcX or 10% FBS for 10 minutes at 4°C, which is very useful for nonspecific binding of LAG-3 in monocytes and dendritic cells. Then stain surface markers CD3, CD8, and LAG-3; after washing, use Foxp3 or Transcription Factor fixation/permeabilization buffer to stain CTLA-4. FMO controls should be performed to determine gating, especially for weak LAG-3 positivity. For every million cells, use a 100 μL system, incubate for 30 minutes at 2–8°C protected from light, and wash twice. Staining CTLA-4 after fixation will not affect surface markers.
