I recommend trying the domestic Hieff Trans 293 (Yeasen) or Vazyme Lipo293. We do lentiviral packaging in Guangzhou and switched from PEI to Hieff Trans; for a 10 cm dish, use 24 µL of reagent for 8 µg of plasmid, change the medium 6–8 h after transfection, and the batch-to-batch fluctuation in packaging titer is much smaller than with self-prepared PEI. In terms of toxicity, the morphology of 293T cells remains essentially unchanged. Within the budget for a 1 mL format, Guangzhou distributors generally have it in stock or can deliver next day. The prerequisite is to dilute it in serum-free basal medium, not directly in serum-containing medium.

If Guangzhou needs delivery within 3 days, prioritize local distributors with stock rather than the brand's official channel. We compared PEIpro with domestic cationic polymers: PEIpro has consistent batch-to-batch performance, but small packaging may exceed your budget; domestic options like PolyJet (SignaGen) sometimes offer suitable pricing and have distributors in Guangzhou. Try it at a 1:3 volume ratio, with 8 μg plasmid corresponding to 24 μL reagent. The key is not simply the brand, but after arrival, using the same batch of 293T cells for small-scale GFP plasmid transfection, checking fluorescence intensity and cell viability at 48 h, and then deciding on the bulk order.

It is not recommended to switch a large amount of reagents all at once. First, fix the PEI source to the commercial PEI MAX 40k (Polysciences powder, freshly prepared), which is low-cost and more stable than laboratory-prepared PEI; if you really want to use a ready-to-use formulation, choose a brand with cold-chain shipping and complete batch-to-batch quality control reports. For a 10 cm dish, 8 μg DNA is on the low side. In my lentiviral system, I usually add 12-16 μg total DNA, and use the transfection reagent according to the reagent instructions rather than applying the PEI ratio. Before ordering from Guangzhou, first ask about inventory and shipping location, and request a batch-specific COA, which can reduce the risk of batch-to-batch variation.

In-house prepared PEI is the most reliable; with imported transfection reagents, extended lead times can halt experiments, and the titer difference is minimal.

It is recommended to prepare both domestic and imported small samples so they can serve as backups when delivery times are unreliable.
