Custom chips with a minimum order of 5 slides are relatively flexible, while those with a minimum order of 10 slides generally have a lower unit price. For screening hybridoma supernatants against 20 antigens, it is advisable not to order too many the first time; 5–10 slides are sufficient for pilot experiments and one batch of supernatants. If more are needed later, you can use the same spotting data to place an additional order. I cannot give you a specific price, but you can break down both quotations into: spotting fee, slide/modification fee, QC fee, and shipping fee, and ask clearly whether reordering waives the spotting setup fee. If the vendor that is 30% more expensive allows a minimum order of 5 slides and has a shorter turnaround time, it may be more cost-effective for early screening, so do not focus only on the unit price.

The sample volume of hybridoma supernatant is usually not a problem. For a single slide without separate barriers, approximately 100–200 μL of supernatant is required; if a multi-well barrier (such as 16 wells) is used, 30–50 μL per well is sufficient. When you need to screen many supernatants, try to have the manufacturer produce chips with barriers or that can be covered with film, so that multiple supernatants can be run on one slide at the same time, greatly increasing throughput. Before loading the sample, centrifuge to remove cell debris, and the culture supernatant can be diluted 1:1 or 1:2. First confirm whether the detection method uses a labeled secondary antibody or directly labels the supernatant, as this will affect the amount used and the background. Ask the manufacturer clearly in advance.

With 20 antigen spots on a single slide, you must ask the manufacturer how many replicate spots are printed for each antigen and how their positions are randomized. Typically, there are 3–4 technical replicates, plus positive and negative control spots. Without replicate spots, it is nearly impossible to determine whether the subsequent supernatant binding signal is real or due to uneven spotting. You can request that replicate spots and controls be marked on the design map, and ask the manufacturer to provide post-spotting scan or quality control data. If screening hybridoma supernatants, it is also recommended to spot a positive control antibody (e.g., anti-His tag) to confirm that the chip spotting and detection workflow is functioning properly.
