Do not ask for a flat all-inclusive price for the entire slide. Have the manufacturer quote separately for dotting processing fees, pathology quality control, HE, sectioning, blank slides, rush service, and tax. For 48+12 cases with 2 replicates per case, the dotting fee is one part, but what really creates the price gap is the depth of quality control: whether only HE is performed or IHC prestaining is added, whether digital scanning is included, and how many blank slides are provided. A two-week rush usually incurs an extra charge. When negotiating, it is advisable to fix the configuration at 120 valid dots, 2 HE quality control slides, and a certain number of blank slides before comparing prices; otherwise, the total prices quoted by different vendors are not comparable. The unit price for multiple dots is generally lower than for small batches, but do not focus only on pushing down the unit price, as rework costs will also be factored in.

Whether delivery within two weeks is feasible depends on the number of blank slides and H&E slides. If it is only chip wax blocks plus a small number of H&E slides, 120 spots including controls, and the wax block quality is acceptable, some manufacturers can do it within two weeks, but the risk is that the archival wax blocks you provide often need to be re-trimmed, and poor embedding can lead to rework. It is recommended that the contract break expedited delivery into milestones such as 7 working days after quality control confirmation, rather than counting from the shipment date. If dozens of blank slides are needed at one time, and they also need to be sectioned first and then baked, the timeline will be even tighter. Expedited fees are usually charged separately by day or by project, so ask clearly whether quality control rework is included.

Before proceeding, have the supplier perform HE quality control on the wax blocks to confirm the tumor cell percentage and the presence or absence of extensive necrosis. The paraffin blocks you provide do not all necessarily qualify for direct punching; many archived blocks have surface oxidation or poor initial fixation, which can lead to section loss and uneven staining in the punched spots. For 48 cases plus 12 adjacent normal cases, with 2 replicate spots per case, that is 120 actual spots, and quality control positions and orientation markers must also be reserved. It is recommended to first ask the other party to evaluate all or some of the blocks using HE sections, and replace or re-label any that fail. In that case, the turnaround time is not simply calculated as spot processing; quality control communication time must also be added.

Require the supplier to provide a draft array design drawing before formal production, indicating cancer spots, adjacent-to-cancer spots, replicate spots, and blank positions, and confirm whether the two replicate spots for each case are adjacent or separated. Specify in the contract that if quality control fails, additional wax blocks can be requested or the work can be redone, to avoid disputes later if staining fails. You can request a pilot batch of a pre-test chip containing 20–30 spots, and perform HE or IHC yourselves to assess spot loss rate and edge spot performance. Before placing a large order, test the supplier's blank slide section thickness uniformity, mounting position deviation, and wax block ID marking—these matter more than promotional PPTs.
