Section detachment is not solely the manufacturer's issue. When edge spots detach during high-temperature retrieval, the common causes are tissue spots being cut too thin, insufficient baking, or the retrieval solution boiling too vigorously. First, confirm whether the sections are 4 μm and whether positively charged slides are used. I typically bake at 60–65°C for at least 2 hours, then bake for another 10 minutes before retrieval; for high-pressure retrieval, use a sealed retrieval box, and allow natural cooling for 20 minutes before opening the lid, which can significantly reduce spot loss. The smaller the spot diameter, the higher the edge proportion, making detachment more likely, so for precious samples, prioritize 1.5 mm or 2.0 mm. Differences among manufacturers mainly lie in array quality control and section replacement services. Before purchasing, ask whether they can provide HE scans for spot integrity rate and same-batch blank sections.

Smaller tissue spots are generally more prone to detachment, but this is not the only factor. For precious samples, 1.0 mm spots are not recommended; 1.5 mm or 2.0 mm margins are more stable. During retrieval, microwave is easier to control than high pressure, but microwave heating is uneven; water bath heat retrieval or low-power microwave can be used, and the buffer should not boil vigorously. FISH tissue detachment may also be related to excessive pepsin digestion. It is recommended to use regular sections first to determine the digestion time, and if necessary, lower the denaturation temperature by 1-2°C. After-sales replacement sections are generally provided by manufacturers, but often at a charge, unless the sample is defective upon arrival. When evaluating cost-effectiveness, do not look only at the unit price; consider the time cost and repeat experiment expenses after tissue detachment.

I have dealt with several domestic suppliers that make TMA slides. It is not accurate to say that any one supplier has no tissue loss at all; the key factors are wax block preparation, microtome blade edge, slide type, and baking parameters. I recommend choosing a manufacturer that has a pathology platform and can perform its own quality control, rather than a company that merely sells chips; such manufacturers are more willing to handle replacement sections. FISH has higher requirements for tissue adhesion because it involves protease digestion and high-temperature denaturation, so silanized slides or positively charged slides should be used, with a section thickness of 3-4 μm, but not too thin. In terms of after-sales service, legitimate manufacturers generally can provide replacement sections for missing spots found upon arrival and for transport-related fragmentation; detachment after staining is more often due to operational factors and is harder to replace free of charge, so this must be confirmed in advance. For an experiment using 10 slides at a time, you can request 2 extra backup slides.

If sections repeatedly detach, it is advisable to confirm the wax melting point, floatation temperature, and baking conditions with the supplier. Some TMA recipient blocks have a relatively low wax melting point, and residual wax during retrieval can affect adhesion, so sections must be thoroughly baked dry. You may switch to APES- or poly-L-lysine-coated slides, but poly-L-lysine may cause slightly higher background. My own workflow is: after sectioning, 37°C overnight or 60°C for 2 hours; during retrieval, use high-temperature-resistant slide racks to prevent sections from sticking to the wall; after IHC retrieval, cool for 20 minutes first; for FISH, try reducing digestion time by 20%–30%. Regarding suppliers, prioritize manufacturers that can provide dot maps and sectioning replacement services. After receiving the goods, first perform one HE stain to check the spot integrity rate, and do not use all 10 slides at once.
