Deep IHC background in liver tissue is not necessarily due to a problem with the antibody itself. F4/80-positive Kupffer cells are distributed in the hepatic sinusoids and are easily confused with stellate cells and endogenous pigment. If you previously used a domestic antibody, there may also be superimposed issues such as secondary antibody species mismatch or biotin interference. It is recommended to switch to a rabbit monoclonal antibody and pair it with a rabbit-specific polymer secondary antibody, avoiding direct staining of mouse tissue with an anti-mouse secondary antibody. For blocking, use 5% normal serum plus 1% BSA, and add 0.05% Tween-20 to the wash buffer. First perform a no-primary-antibody control to confirm whether the background comes from the primary antibody or from endogenous enzymes or biotin, then decide which step to change.

When choosing imported aliquoted antibodies on a limited budget, make sure the supplier provides the clone number and original lot number. Only clones that can be verified in the literature, such as SP115, D2S9R, or BM8, are reliable. For paraffin-embedded liver tissue, I recommend SP115 or D2S9R. BM8 has relatively average sensitivity on paraffin sections, and many studies using it are based on frozen sections. For dilution, I suggest first performing titration at three levels: 1:100, 1:200, and 1:400, with EDTA pH9.0 used uniformly for antigen retrieval. A weekly volume of 20 slides is not large. After aliquoting the formal antibody, store it at -20°C and avoid repeated freeze-thaw cycles. When switching to a new antibody, stain it together with the same batch of positive liver slides to check whether the background and positive intensity remain stable.

In terms of literature support, the rabbit monoclonal antibodies Abcam SP115 and CST D2S9R are commonly used for F4/80 in paraffin-embedded mouse liver, with clear chromosomal cellular localization and perinuclear staining that is not particularly blurry. Be careful not to use citric acid at low pressure for antigen retrieval, otherwise the F4/80 membrane/cytoplasmic signal tends to be weak and appears like background. Control DAB development to 3-5 minutes, counterstain lightly with hematoxylin, as liver tissue cytoplasm itself readily adsorbs brown-yellow color. If the laboratory already has an imported secondary antibody system, only the primary antibody needs to be replaced, making costs more controllable. Before purchasing, check whether the antibody datasheet includes IHC-P species and liver tissue application images.

For mouse liver paraffin F4/80, prioritize the clone number rather than just the brand. I am currently using CST's F4/80 (D2S9R) XP rabbit monoclonal antibody, catalog number 70076, with EDTA pH 9.0 pressure cooker antigen retrieval for 2 minutes, 1:200 overnight at 4°C, and the background is much cleaner than the domestic rat monoclonal antibody I used before. For liver tissue, first inactivate endogenous peroxidase with 3% H2O2 for 15 minutes, then block with 5% normal goat serum at room temperature for 30 minutes. A full-size vial is generally enough within a budget of 2500, and it is recommended to first get a small pack or trial sample from a distributor, stain a few positive controls, and then buy in bulk.
