We have expressed His tag protein in BL21 (DE3) using two domestic and Sigma methods, at 0.5 mM and 16 ° C overnight. The intensity of the target band on SDS-PAGE was basically the same, and the difference in Ni column elution yield was also within an acceptable range. There may be greater fluctuations between different domestic manufacturers and batches. It is recommended to fix the supplier and batch number. You can produce 20 bottles at a time. Before changing, take 2-3 bottles of the new batch and compare them with the imported batch to see the total protein and supernatant. The delivery time for domestic products is generally 3-7 days, and the price is much cheaper per gram compared to imports. Specific inquiries need to be made, and there are significant differences in channels.

It is not advisable to focus only on the brand; check COA purity, moisture, and other parameters. The induction effect of IPTG is mainly determined by the free effective concentration. If domestic and imported products meet conventional specifications in purity, their induction efficiency generally will not differ much. You may request the supplier to provide a COA, focusing on purity, loss on drying, and heavy metals. After arrival, inspect the appearance for caking and color. For large packages, aliquot and store dry first. Before changing batches, use the same batch of competent cells and the same batch of culture medium to perform small-scale induction validation and compare the target protein yield; do not directly scale up to 20 bottles. In terms of price, imported products are usually more expensive, while domestic 25 g has a lower price per gram, but the validation cost must be included.

You use 20 bottles of 200 mL at a time, and the consumption of IPTG is relatively high. If it is verified to be OK, domestic production is indeed more suitable. When we switch to domestic products, we first buy small packages for small-scale testing, confirm the induction efficiency, and directly lock 25g. Generally, IPTG is not an expression bottleneck, and there is not much difference between imported and qualified domestic products under overnight conditions of 0.5 mM at 16 degrees Celsius. Pay attention to the short delivery time of domestic products, but it is best to compare them with imported ones after arrival to avoid batch fluctuations. Don't just look at the quotation for price, imports may include shipping and customs duties, and there are many domestic spot goods. Please consult the regional agent for details. We generally require samples with the same batch number to be retained for easy traceability.

Changes in inducer batches can directly affect yield, so it is recommended to establish an incoming material verification process. For each new batch, perform a parallel control using the same plasmid, the same competent cells, and the same induction conditions, then compare band grayscale by SDS-PAGE scanning or ImageJ; if the difference is within an acceptable range, it can be used. Most domestic and imported products show little difference in BL21(DE3), but some domestic batches differ in pH or color after dissolution; if any abnormality occurs, stop using them immediately. Domestic products do have advantages in price and lead time, usually in stock within 1 week; imported products may take 2-4 weeks. For your usage of 20 bottles, it is recommended to purchase a sufficient quantity at one time and fix the batch to reduce switching frequency.
