IgG levels in regular fetal bovine serum are typically lower than those in newborn or adult bovine serum, but there can be considerable variation between brands and lots. If downstream purification uses Protein A/G, bovine IgG can indeed compete with your monoclonal antibody for binding to the resin, causing reduced capacity, background bands, or interference with subsequent validation. If you are only performing ELISA or functional primary screening during the hybridoma screening stage, regular serum is generally acceptable; however, if you will soon move into purification, it is advisable to prioritize low-IgG or antibody production-specific serum, or switch to serum-free/low-IgG medium at the mid-to-late stage. Do not rely solely on the product name; ask the supplier to provide IgG residual data for that lot and perform an spike-in purification validation yourself, which carries lower risk than simply switching to regular serum.

When low-IgG supply is unstable, rather than searching everywhere for spot stock, it is better to find an agent that can sign a quarterly or semi-annual stocking agreement. In terms of brands, you can compare imported Gibco, Hyclone, and Biowest with leading domestic brands such as ExCell and SiJiQing, but the key is not reputation; it is whether the low-IgG product line regularly has stock on hand, whether batch quality inspection certificates can be provided, and whether small-sample trial batches are accepted. Based on your budget, there is a relatively wide range of options for regular fetal bovine serum, while the low-IgG dedicated line may be tighter, so it is recommended to prioritize spending your budget on locking in the same lot number and reducing lot switching. A lead time within two weeks is usually achievable: imported spot stock takes 1-2 weeks, and domestic is faster. Before ordering, confirm whether it is the same lot number and whether IgG and endotoxin data are provided.

Fetal bovine serum mainly provides adherent/semi-adherent support, lipids, and growth factors for hybridomas, and can also affect the stability of antibody secretion. When evaluating lots, it is recommended to examine: cell doubling time, peak viability, target antibody titer in the culture supernatant (ELISA), cell morphology, and clumping. A volume of 2-3 bottles per month is not large, and the greatest concern is frequent lot switching. It is recommended to have the supplier send 3 candidate lot numbers at 50-100 mL each as small samples at one time, revive the same hybridoma line uniformly, and perform a 3-5 day comparison of growth and secretion. After locking in 1-2 lot numbers, require the distributor to keep them in stock. Focus on endotoxin below 10 EU/mL, low hemoglobin, and stable pH; serum that is too dark or reddish often indicates possible problems in the collection process.
