Firstly, investigate the operational steps: if PBS containing calcium and magnesium is used during reconstitution, trypsin activity may be affected; Excessive reconstitution temperature or forceful vortexing can also reduce activity. After being packaged and stored at -20 ℃, if the same tube is repeatedly frozen and thawed more than twice, the activity decreases significantly. Suggest using pre cooled calcium free magnesium PBS for reconstitution, gently mix on ice, and divide into small volumes for single use to avoid repeated freeze-thaw cycles. If the packaging volume is large and the remaining portion is frozen after each use, it may be extended from 3 minutes to 8 minutes, which is more like a storage issue rather than a batch issue.

To assess batch-to-batch variability, it is recommended to compare an unopened vial of lyophilized powder from the same batch with a previously retained sample. If a newly opened vial from the same batch also shows a significantly longer digestion time after reconstitution, it indicates that the batch itself may have lower activity; if a newly opened vial from a different lot number performs normally, then improper storage after reconstitution is the most likely cause. You can request the Certificate of Analysis (COA) for that batch from the manufacturer to verify the activity unit range. Note that vial-to-vial variability can also exist within the same batch, especially when the aliquoting process is inconsistent. Two vials from the same batch can be reconstituted simultaneously to rule out chance variation.

BAEE and USP are not a simple conversion relationship. BAEE is usually used as a substrate to determine esterase activity, while USP units are defined by pharmacopoeial methods. Different detection methods have different substrates and conditions, and cannot be directly converted to a fixed multiple. It depends on which unit is marked on the manufacturer's COA, and sometimes it may be provided simultaneously. If you test BAEE activity yourself, you need to benchmark it against the manufacturer's method, temperature, pH, and substrate concentration, otherwise the data cannot be compared. Do not use digestion time to infer units, as cell digestion is also influenced by EDTA, pH, and cell type.

If it has not expired but the activity is suspected to be below specification, you may apply for a replacement, but evidence is required. Retain an unopened sample from the same lot, and keep reconstitution records including date, buffer, procedure, aliquoting, and freeze-thaw cycle records. It is best to provide activity assay data or a parallel comparison with a normal lot. Contact the supplier or the manufacturer's QC and request retesting of the retained sample. If their retest also deviates from the COA range, they will generally offer a replacement or refund; if the retest passes, they may consider it a storage or handling issue. It is recommended to first perform a standardized reconstitution internally and measure enzyme activity before initiating a complaint, which will make communication smoother.
